In general, how often do you have to drain a 16oz canister? Is it big enough to last between oil changes?

respectfully disagree. Depending on climate where you live/drive, these engines will typically fill 12-16 oz in 3000-5000 miles

Is that why brands like Rx offer a 32oz can? I'm assuming if the can is too small and overfills, that's really bad because the incoming and out going fluid has no where to go but into your engine? Or is there some kind of overflow protection?

Well, fuel in oil is a huge ecoboost problem and condensate buildup in the intercooler is a HUGE, life endangering problem.
