Not all tire manufacturers follow the same exact rules for this, some don't even bother putting the dot on, others have multiple dots marking both the heaviest and lightest part of the tire, but most use the yellow dot to be lined up with the valve stem, the only time you should bother with the red dot is if your wheels are marked for their heaviest point, and then you should try to match your tires to your wheels so the yellow dot is as close as possible to the valve stem and the red dot is as close as possible to the heavy marking on your wheel
